? ?“Lifespan extension by novel chromatin-regulated stress response pathways - New lessons learned from the budding yeast”

报告摘要: In this presentation, I will talk about two recent projects from my laboratory, both of which were carried out in the budding yeast but will have implications in all eukaryotes. In one project, we studied the effects of histone dosage on lifespan and discovered a novel form of stress response, Chromatin Architectural Defect (CAD) response, which when activated by moderate chromatin stress could extend lifespan. We found that CAD response was regulated by stress response transcription factor Gis1 and its longevity effect was mediated through TOR inhibition. Furthermore, we have evidence that CAD response may be evolutionarily conserved. In the other project, we performed an unbiased high throughput lifespan screen for the ORF deletion library and discovered that a class-II histone deacetylase, the HDA complex, regulated aging through its target of H3K18ac. Interestingly, HDA specifically targets the trehalose and glycogen metabolic pathways, which play critical roles in stress response, ranging from yeast, to plants and invertebrate animals. Furthermore, we found that this longevity effect of HDA was independent of the Cyc8 (Ssn6) – Tup1 repressor complex that has been previously shown to interact with HDA and coordinate transcriptional repression. We found that losing HDA activity rendered cells in a poised state in response to stress, giving them an advantage in responding age-associated stresses, hence promoting longevity.
